Colonel Glenn Road runs mostly east-to-west from its intersection with Asher Avenue near the campus of the University of Arkansas at Little Rock to its dead end west of Ferndale-Congo Road in rural Pulaski County. Part of the eastern end of the road is designated as Arkansas Highway 5 before following Stagecoach Road southwest out of the city of Little Rock.
Colonel Glenn Road intersects with a number of major roads, including University Avenue, Stagecoach Road, Shackleford Road, Interstate 430, Burlingame Road, County Line Road, and Ferndale-Congo Road.
