The first local Walgreen Drug Store was located at 501 Main Street in downtown Little Rock. Walgreen's opened at the location in the 1930s.

During the 1960s Civil Rights struggle the store experienced sit-ins by African Americans protesting the store's segregated lunch counters. The store closed on January 15, 1977, and was replaced by a Country Kitchen chicken franchise.
