In central Arkansas the route passed through Old Austin, Parnell, Jacksonville, North Little Rock, Little Rock, Oak Grove, Morgan, Palarm, Mayflower, Conway, Cadron Settlement, Menifee, Plumerville, Morrilton, Kenwood, and Blackwell.

The general stage office was located at Anthony House in Little Rock. A Butterfield stage landing was located on the Arkansas River near Toad Suck.
